    Longitudinal vortices in a transitioning boundary layer

    Naturally occurring spanwise variations of the streamwise velocity component, characteristic of longitudinal vortices embedded in a transitioning boundary layer were explored using hot-wire anemometers. A vibrating ribbon introduced stable or unstable Tollmien-Schlichting waves into the laminar boundary layer. These damped or growing disturbances always developed a strong three dimensional pattern even though no spanwise perturbations were artificially induced. Changing the radius of the leading edge and other modifications to the flat plate, wind tunnel and boundary layer did not alter the spanwise wavelength of the vortices

    Inflectional instabilities in the wall region of bounded turbulent shear flows

    The primary thrust of this research was to identify one or more mechanisms responsible for strong turbulence production events in the wall region of bounded turbulent shear flows. Based upon previous work in a transitional boundary layer, it seemed highly probable that the production events were preceded by an inflectional velocity profile which formed on the interface between the low-speed streak and the surrounding fluid. In bounded transitional flows, this unstable profile developed velocity fluctuations in the streamwise direction and in the direction perpendicular to the sheared surface. The rapid growth of these instabilities leads to a breakdown and production of turbulence. Since bounded turbulent flows have many of the same characteristics, they may also experience a similar type of breakdown and turbulence production mechanism

    Effects of Calcium, Strontium, and Magnesium on the Coccolithophorid Cricosphaera (Hymenomonas) carterae. I. Calcification

    The present investigation concerns the effects of calcium, strontium, and magnesium on calcification and mineralogy of the calcified bodies (coccoliths) of the coccolithophorid Cricosphaera (Hymenomonas) carterae. The capacity of cells to calcify in various concentrations of these ions was examined following preliminary decalcification in CO2. At a concentration of 10-2 M Ca, 75% of the cells formed coccoliths within 24 h and almost all cells were recalcified after 2 days. At 10-3 or 10-4 M Ca no recalcification occurred. However, with the addition of Sr to the Ca-deficient media, calcification took place as shown by observations of coccoliths and by analysis of Ca. The percentage of calcified cells increased with increasing concentrations of Sr. Strontium added to a Ca-deficient media was much more effective than an equivalent concentration of Ca. No Sr was deposited in the coccoliths. X-ray analysis demonstrated that calcite was deposited by cells in all concentrations of Ca and Sr at which calcification took place. At concentrations of Mg in the media from 0.0 to 4.2×10-2 M, the cells retained their ability to calcify, although calcification was markedly reduced in the absence of Mg. In low Mg concentrations (1.3×10-4 and 4.2×10-6 M), the coccoliths were 60% calcite and 40% aragonite, and in the absence of Mg, only calcite was formed

    Current interest in utilization of coccoliths for paleoclimate reconstruction necessitates background information on environmental limits for growth and coccolith production as well as examination of cell ultrastructure in specimens collected in the field and in cultured representatives. Successful isolation of the two geologically important species Gephyrocapsa oceanica (strain A674) and Cyclococcolithina leptopora (strain A650) allows investigation of ultrastructure in cultured forms. Fine structure of cells and coccoliths was observed in the SEM using critical point dried preparations and ultrastructure was examined with the transmission electron microscope. Coccoliths are formed intracellularly and appear to form within Golgi-derived vesicles located near the nuclear membrane. Formation and development of coccoliths in the two species resemble these processes in Emiliania huxleyi but differ from those of Cricosphaera carterae, notably in the absence of coccolithosomes and scales and in the fact that coccoliths are produced intracellularly one at a time

    Initiation of Turbulent Spots in a Laminar Boundary Layer by Rigid Falling Particulates

    A transitional laminar boundary layer is developed on a 1m wide km long flat plate in a 0.6m deep water channel with a freestream velocity of 15-50 cm/s. A particulate dispenser under computer control ejects individual particles having diameters of 1/3 delta into the free stream. The particulates are introduced with an initial velocity of U(sub infinity) in the direction of the free stream. They have differing specific gravities of 1.03-2.7 which introduces an additional non-dimensional parameter relating the time taken to traverse the boundary layer to the convective time scale. The particulates produce a wake in the upper region of the boundary layer as they sink towards the wall. Visualization data taken over the range 5 x 10(exp 4) less than Re(sub x) less than 5 x 10(exp 5) indicate that turbulent spots are produced by the disturbances due to the wake rather than by the particulates themselves. This suggests that the spot formation process in this case may be inviscid in nature and may not be strongly influenced by the presence of the wall

    Boynton-Delray Coastal Water Quality Monitoring Program

    This report discusses a sequence of six cruises in the vicinity of the Boynton-Delray (South Central) treated-wastewater plant outfall plume (26°27\u2743 N, 80°2\u2732 W), the Boynton Inlet (26°32\u2743 N, 80°2\u2730 W), and the Lake Worth Lagoon, Palm Beach County, Florida. The sampling cruises took place on June 5-6, 2007; August 28-29, 2007; October 18-19, 2007; February 14 and 18, 2008; May 19-20, 2008; and July 11-13, 2008. Water was sampled at 18 locations at the surface, middle, and near the seafloor (where there was sufficient depth) for a total of 45 samples; these samples were analyzed for a variety of nutrients and related parameters. The water sampling unit contained a conductivity-temperature-depth (CTD) instrument from which data were obtained at each sampling site. Synchronal ocean current data were measured by a nearby acoustic Doppler current profiler (ADCP) instrument. The inlet measurements were consistently lower in salinity and more acidic (lower in pH) than the coastal ocean and were warmer during the May and, especially, during the February cruises. For most analytes, viz., nitrite+nitrate (N+N), total suspended solids (TSS), chlorophyll-a, silica (Si), and total dissolved nitrogen (TDN), the lagoon concentrations were significantly higher than the coastal ocean; the inlet concentrations appeared to be consistent with lagoon water with partial mixing with the coastal ocean, as expected. Estimates of the nutrient flux to the coastal ocean were computed: approximately 1,500 kg of dissolved nitrogen (N), 2,350 kg of silicate (Si), 33 kg of orthophosphate (P), and 59 kg of ammonium (NH4) per day were delivered to the coastal ocean through the inlet. The outfall boil at South Central outfall (the smallest in volume of the six outfalls in southeast Florida) is only visible under ideal conditions. In the six cruises described in this document, the outfall boil could be found in only one cruise (August 28-29, 2007). Elevated concentrations of nutrients (N+N, P, Si, and P) at the outfall vicinity were measured, and these concentrations decreased rapidly away from the outfall for most analytes, to become undistinguished from the background within 3 km or less. Not finding the boil, however, in five of six cruises meant that the waters with the highest concentrations were probably missed. When the boil was sampled in August 2007, N+N, P, and total dissolved phosphorus (TDP) concentrations at the boil were roughly the same as from the inlet. For other analytes (chlorophyll-a, TSS, Si, and dissolved organic carbon [DOC]), the concentrations at or near the outfall were significantly less than those from the lagoon and inlet on most of the cruises. The coastal ocean appeared to be significantly impacted by the Boynton Inlet and less so from the inlet. A suggestion of a source to the south was seen in some analytes. Measurements from the Gulf Stream Reef area were the lowest in the study, and may provide “background” concentrations for this region. As expected, the coastal ocean was warmer and more stratified in the summer compared to the winter, e.g., whereas no thermocline was noted in the CTD data from February 2007, a strong thermocline was observed in most casts during July 2008. In certain cases (e.g., N+N in June 2007, pH in July 2008), an increase in the concentration (decrease for pH) from north to south implied a source from the south, e.g., the Boca Raton Inlet or Boca Raton outfall

    Animal-related factors associated with moderate-to-severe diarrhea in children younger than five years in western Kenya: A matched case-control study

    Background Diarrheal disease remains among the leading causes of global mortality in children younger than 5 years. Exposure to domestic animals may be a risk factor for diarrheal disease. The objectives of this study were to identify animal-related exposures associated with cases of moderate-to-severe diarrhea (MSD) in children in rural western Kenya, and to identify the major zoonotic enteric pathogens present in domestic animals residing in the homesteads of case and control children. Methodology/Principal findings We characterized animal-related exposures in a subset of case and control children (n = 73 pairs matched on age, sex and location) with reported animal presence at home enrolled in the Global Enteric Multicenter Study in western Kenya, and analysed these for an association with MSD. We identified potentially zoonotic enteric pathogens in pooled fecal specimens collected from domestic animals resident at children’s homesteads. Variables that were associated with decreased risk of MSD were washing hands after animal contact (matched odds ratio [MOR] = 0.2; 95% CI 0.08–0.7), and presence of adult sheep that were not confined in a pen overnight (MOR = 0.1; 0.02–0.5). Variables that were associated with increased risk of MSD were increasing number of sheep owned (MOR = 1.2; 1.0–1.5), frequent observation of fresh rodent excreta (feces/urine) outside the house (MOR = 7.5; 1.5–37.2), and participation of the child in providing water to chickens (MOR = 3.8; 1.2–12.2). Of 691 pooled specimens collected from 2,174 domestic animals, 159 pools (23%) tested positive for one or more potentially zoonotic enteric pathogens (Campylobacter jejuni, C. coli, non-typhoidal Salmonella, diarrheagenic E. coli, Giardia, Cryptosporidium, or rotavirus). We did not find any association between the presence of particular pathogens in household animals, and MSD in children. Conclusions and significance Public health agencies should continue to promote frequent hand washing, including after animal contact, to reduce the risk of MSD. Future studies should address specific causal relations of MSD with sheep and chicken husbandry practices, and with the presence of rodents

    Statistical Methods in Recent HIV Noninferiority Trials: Reanalysis of 11 Trials

    Background: In recent years the ‘‘noninferiority’ ’ trial has emerged as the new standard design for HIV drug development among antiretroviral patients often with a primary endpoint based on the difference in success rates between the two treatment groups. Different statistical methods have been introduced to provide confidence intervals for that difference. The main objective is to investigate whether the choice of the statistical method changes the conclusion of the trials. Methods: We presented 11 trials published in 2010 using a difference in proportions as the primary endpoint. In these trials, 5 different statistical methods have been used to estimate such confidence intervals. The five methods are described and applied to data from the 11 trials. The noninferiority of the new treatment is not demonstrated if the prespecified noninferiority margin it includes in the confidence interval of the treatment difference. Results: Results indicated that confidence intervals can be quite different according to the method used. In many situations, however, conclusions of the trials are not altered because point estimates of the treatment difference were too far from the prespecified noninferiority margins. Nevertheless, in few trials the use of different statistical methods led to different conclusions. In particular the use of ‘‘exact’ ’ methods can be very confusing.
